I was put on this combo in Nov. In Dec, my bloodwork showed dangerously low white blood cell count. My LLMD called on a Sat to tell me to quit taking all abx.
In Jan, my WBC count was good again, so doc had me start the rifampin only back up. He suspects it was the combo that pushed my WBC so low.

My 3 year old and 5 year old are on this combo, plus Mepron too. They are doing well on it.
Our LLMD said that Rifampin should be paired with another drug that hits bart, such as zith, biaxin, bactrim, etc. Otherwise, it could become resistant to the bart and stop working.

Yes, they interact- you can read more at http://www.drugs.com/drug_interactions.php however, many LLMD's have decided that the amount of interaction isn't that important- you still get the benefits of having two drugs hit the same pathogen, it's just the exact dosage level that varies a bit.

I once read something here about how someone's LLMD had a specific way he suggested for taking these two meds- something like take one with food, then wait a certain amount of time, then take the other. It made a lot of sense pharmacologically at the time but I was not on the drugs then. Does that ring a bell for anyone? Could you shed some light on this?
