Looking it up on Wikipedia and I don't see anything about it being an anti-microbial.
I see some info on neuroprotection and as an antioxidant.
Is this why people take it, or is there some substantiated claim for it as an anti-spirochetal herb?

The sheet my doctor gave me (taken from Healing Lyme and/or Healing Lyme Disease Naturally) states that its benefits (when using a formulation that specifically contains polygonum root extract not just resveratrol) are: 1) Contains Japanese Knotwood that is active agains a number of gram-negative and gram-positive bacteria, is anti-inflammatory for both arthritic and bacterial inflammation and helps reduce Herxeimer reactions. 2) Polygonum specifically protects the brain from inflammatory damage, microbal endotoxins, and bacterial infections. 3. Useful for spirchete infection and bartonella and other coinfections.
