Has anybody else seen this?

http://www.medpagetoday.com/ProductAlert/Prescriptions/tb/3263

It looks like grapefruit juice also interferes with the P450 3A4 pathway (just like diflucan, although to a lesser degree I suppose). Has anyone seen a positive effect with grapefruit juice?

up.
All this microbiology goes right over my head, but a dear friend has been put on Diflucan for Lyme. Would love to hear how others are doing on it.

I found this at
http://rxlist.com/cgi/generic/flucon_cp.htm
[quote]
CLINICAL PHARMACOLOGY

Mode of Action

Fluconazole is a highly selective inhibitor of fungal cytochrome P-450 sterol C-14 alpha-demethylation. Mammalian cell demethylation is much less sensitive to fluconazole inhibition. The subsequent loss of normal sterols correlates with the accumulation of 14 alpha-methyl sterols in fungi and may be responsible for the fungistatic activity of fluconazole.[end quote]

I decided not to take the diflucan

I feel so congested (intestines) when I take it and I believe it makes my constipation worse.

Also I just feel so crummy.
This might be a herx,

But Im also herxing hard on just 50-60 mg of DOXY.
So all taken with stride.

Drinking lots of water and taking colon cleanse product for constipation.

Interestinglly Flagyl cleared up some constipation but now its even rearing back.

Worked out 20 minues yesturday and then sauna.
So much pain and FOG afterwards.

Fog and pain, lack of sleep, tossing turning, postnasal drip, vertigo, bruning pains.

Thats all the symptoms Im left with.

My opinion is that I have different bugs in me.
They all do better when I feel worse so thats where they try to keep and convince me.

Diflucan is out for now.
Also had side effects from the biaxin while on just 1/2 diflucan.

My P450 enzyme system gets clogged quically with this.

All the studie show that the way it kills yeast is to poison it to death by clogging its P450 pathway for elimination of cellular waste.

Hmm when does the benefit outweigh the risk.

Water Water Water
Positive thoughts
DOXY burning and fatigue.

Herbs for energy as needed and while they work.

Less rest more play and motivation.

More rest does me no GOOD!!!!

Thanks for a chance to rant***********

I recently had the genomics test done. It showed that I also have trouble with the P450 pathway, shows that I can't take diflucan or flagyl because I have trouble getting estrogen out of my body and these meds block that process

Interesting because I have noticed that when I take long term diflucan or flagyl (over one month) My boobs get bigger and sore. I couldn't figure that out. All the doctors would just look at me and say "I have never heard this"

Well my husband was loving it but I wasn't

It also showed that I am mising a glatathoine gene! I only have 2 not 3! No wonder I have neuro lyme with elevated mercury levels.

May I ask how much this test costed and was it covered by insurance?

I just got billed $956 for test that was performed last year. It was a CD-57 test for an NK count.

Go figure! Insurance said they wont cover it, my father just keeps sending it back and telling them to go to hell.

He helps witht the bills.

Btw,
P450 interference was asserted for milk thistle also:
http://www.ncbi.nlm.nih.gov/entrez/query.fcgi?cmd=Retrieve&db=PubMed&list_uids=11038151&dopt=Abstract

This research paper from 2000 has been quoted at several places.

But another research paper from 2004/5 says (concerning milk thistle) that p450 interference doesn't happen in vivo:
