Hi Guys, I would like advice from anyone who has taken Tini at a higher dose than 1g/day? What dose did you take, how long did you take it for and how beneficial was it!!? I would like to try this but I'm concerned!
Lyme patients are usually prescribed a maximum of 500mg twice daily (b.i.d) (1g/day), but I believe a benefit for some, could be gained by increasing this dose - but would an increased dose over 1g/day sustained for weeks or months be safe for the patient?

Regarding lyme effectiveness, Eva Sapi conducted this great in-vitro (out of the body) research in 2011 which shows tini, in this instance, kind of out in front. See particularly the results text and the graphs towards the end of this article.
Regarding lyme higher doses Brorson's research says: " Our results show that TZ as a single agent is not sufficient to treat Borrelia infections because mobile spirochetes are highly resistant to this agent. However, TZ has the ability to inhibit the development of cystic forms and to disrupt spirochetes and core structures inside the cysts at concentrations achievable in vivo by the administration of a single 1.5-g dose [3]."
Reading the 'Results and Discussions' section for the above, they do emphasise dosage regarding effectiveness quite clearly.

I took it as part of a number of different combos, over several months, I felt they were helpful and that it played its role in the combos. It was always 750mg twice a day (total 1500mg/day).

Michael - Thanks for your higher dosage - very interesting.
I noticed this: http://www.rxlist.com/tindamax-drug/indications-dosage.htm "Amebic Liver Abscess: The recommended dose in adults is a 2 g dose per day for 3-5 days taken with food. In pediatric patients older than three years of age, the recommended dose is 50 mg/kg/day (up to 2 g per day) for 3-5 days with food. There are limited pediatric data on durations of therapy exceeding 3 days, although a small number of children were treated for 5 days without additional reported adverse reactions. Children should be closely monitored when treatment durations exceed 3 days."
Anyone else taken a higher dose than 1000mg / 1g per day of tini?

Some more info on TZ: Pfizer the main manufacturer directs here for data on the drug EMC
says: "Fasigyn is rapidly and completely absorbed following oral administration. In studies with healthy volunteers receiving 2g tinidazole orally, peak serum levels of 40-51 micrograms/ml were achieved within two hours and decreased to between 11-19 micrograms/ml at 24 hours. Healthy volunteers who received 800mg and 1.6g tinidazole IV over 10-15 minutes achieved peak plasma concentrations that ranged from 14 to 21mcg/ml for the 800mg dose and averaged 32mcg/ml for the 1.6g dose. At 24 hours post-infusion, plasma levels of tinidazole decreased to 4-5mcg/ml and 8.6mcg/ml respectively, justifying once daily dosing."
Also seem safe - BUT REGULAR H&B Blood monitoring should be done while taking this drug - always seek proper medical advice while taking this drug!:
"� Use in Renal Impairment Dosage adjustments in patients with impaired renal function are generally not necessary. However because tinidazole is easily removed by haemodialysis, patients may require additional doses of tinidazole to compensate.1,2� Tinidazole is excreted by the liver and kidneys. Studies in healthy patients have shown that over 5 days, 60-65% of an administered dose is excreted by the kidneys with 20-25% of the administered dose excreted as unchanged tinidazole. Up to 5% of the administered dose is excreted in the faeces.3 Studies in patients with renal failure creatinine clearance <22ml/min indicate that there is no statistically significant change in tinidazole pharmacokinetic parameters in these patients. See section 4.2 � Posology and Method of Administration."

I found some very good info here back from 2005 for anyone taking or considering taking Tini
Please add comments about the above link in that thread and only post in here about info on taking tini in DOSES ABOVE 1g PER DAY ( i.e. 2 x 500mg per day)
